Depends where you're going.

For me it'd be check the oil, tyres and fluids then pedal to the metal at 3am, hit the high road for Scotland and don't stop till I'm past Glasgow.

For most others I guess it'd be make sure you have your passport/visa/tickets, travel money or cards sorted, and remember the Imodium
